Biochemical Characterization of Histone Variants and Post-translationally Modified Nucleosomes
Project Description
Eukaryotic genomic DNA is packed by nucleosomes that consist of histone octamers and 146 bp DNA in eukaryotic cells. The goal of this project is to quantitate the stability of nucleosomes that contain histone variants and post-translationally modified histones using biochemical methods.

Applicant's Roles
Expressing the histone proteins in E. coli., and purifying them using FPLC and/or HPLC. After the purification, nucleosomes will be reconstituted in vitro. With the reconstituted nucleosomes, analytical ultracentrifugation will be used for measuring the stability of nucleosomes. Students will work with other lab members in a collaborative way.

Students will learn to how to approach research problems with an analytical approach and develop scientific thinking skills. Students will receive training in modern biochemical and molecular methods through the project.
